Haven't used the Nook much (just briefly at Barnes & Noble), but from what I've seen I very much prefer my Kindle. The color screen would be nice for some things, but the smaller Kindle more easily fits in your pocket/coat/bag and the month-long battery life far surpasses the 6-8 hours of the Nook.

My instinct is that overall, the Kindle is better for novel readers and the color Nook is better for magazine/comic book readers who would do well to have the larger screen and full color display.
